    On the Systematic Errors of Cosmological-Scale Gravity Tests using Redshift Space Distortion: Non-linear Effects and the Halo Bias

    Redshift space distortion (RSD) observed in galaxy redshift surveys is a powerful tool to test gravity theories on cosmological scales, but the systematic uncertainties must carefully be examined for future surveys with large statistics. Here we employ various analytic models of RSD and estimate the systematic errors on measurements of the structure growth-rate parameter, fσ8f\sigma_8, induced by non-linear effects and the halo bias with respect to the dark matter distribution, by using halo catalogues from 40 realisations of 3.4×1083.4 \times 10^8 comoving h3h^{-3}Mpc3^3 cosmological N-body simulations. We consider hypothetical redshift surveys at redshifts z=0.5, 1.35 and 2, and different minimum halo mass thresholds in the range of 5.0×10115.0 \times 10^{11} -- 2.0×1013h1M2.0 \times 10^{13} h^{-1} M_\odot. We find that the systematic error of fσ8f\sigma_8 is greatly reduced to ~5 per cent level, when a recently proposed analytical formula of RSD that takes into account the higher-order coupling between the density and velocity fields is adopted, with a scale-dependent parametric bias model. Dependence of the systematic error on the halo mass, the redshift, and the maximum wavenumber used in the analysis is discussed. We also find that the Wilson-Hilferty transformation is useful to improve the accuracy of likelihood analysis when only a small number of modes are available in power spectrum measurements.Comment: 10 pages, 8 figures, 1 table, accepted for publication in MNRA

    Existence and uniqueness for Legendre curves

    We give a moving frame of a Legendre curve (or, a frontal) in the unite tangent bundle and define a pair of smooth functions of a Legendre curve like as the curvature of a regular plane curve. The existence and uniqueness for Legendre curves are holded like as regular plane curves. It is quite useful to analyse the Legendre curves. As applications, we consider contact between Legendre curves and the arc-length parameter of Legendre immersions in the unite tangent bundle.

    The Subaru FMOS Galaxy Redshift Survey (FastSound). II. The Emission Line Catalog and Properties of Emission Line Galaxies

    We present basic properties of \sim3,300 emission line galaxies detected by the FastSound survey, which are mostly Hα\alpha emitters at zz \sim 1.2-1.5 in the total area of about 20 deg2^2, with the Hα\alpha flux sensitivity limit of 1.6×1016erg cm2s1\sim 1.6 \times 10^{-16} \rm erg \ cm^{-2} s^{-1} at 4.5 sigma. This paper presents the catalogs of the FastSound emission lines and galaxies, which will be open to the public in the near future. We also present basic properties of typical FastSound Hα\alpha emitters, which have Hα\alpha luminosities of 1041.810^{41.8}-1043.310^{43.3} erg/s, SFRs of 20--500 MM_\odot/yr, and stellar masses of 1010.010^{10.0}--1011.310^{11.3} MM_\odot. The 3D distribution maps for the four fields of CFHTLS W1--4 are presented, clearly showing large scale clustering of galaxies at the scale of \sim 100--600 comoving Mpc. Based on 1,105 galaxies with detections of multiple emission lines, we estimate that contamination of non-Hα\alpha lines is about 4% in the single-line emission galaxies, which are mostly [OIII]λ\lambda5007. This contamination fraction is also confirmed by the stacked spectrum of all the FastSound spectra, in which Hα\alpha, [NII]λλ\lambda \lambda6548,6583, [SII]λλ\lambda \lambda6717, 6731, and [OI]λλ\lambda \lambda6300,6364 are seen.Comment: 17 pages, 15 figures, accepted for publication in PAS

    Expression of 5-Hydroxytryptamine Receptors in Human Urinary Bladders with Benign Prostatic Hyperplasia

    Introduction: This study investigated the mRNA expression pattern and distribution of 5-hydroxytryptamine (5-HT) receptors 5-HT2A, 5-HT2B, 5-HT3A, 5-HT4, and 5-HT7 within the urothelium and detrusor of normal bladder tissue and in the urothelium of bladders from patients with benign prostatic hyperplasia (BPH). Methods: Normal urinary bladder specimens were obtained from 13 patients undergoing radical cystectomy due to bladder cancer (normal group) and BPH specimens were obtained from 27 benign prostatic obstruction patients receiving transurethral prostatectomy or retropubic prostatectomy. Receptor subtype mRNA expression was determined by real-time reverse transcription polymerase chain reaction on urothelium, detrusor, and whole mucosal preparations. Receptor distribution was determined by immunohistochemistry. Results: In normal tissues, expressions of 5-HT2B and 5-HT7 receptor mRNAs in the urothelium, detrusor, and whole mucosa were greater than the average expression for all receptor subtype mRNAs. 5-HT2B receptor protein was distributed in the apical urothelium and among the detrusor smooth muscle layers. In contrast, the 5-HT7 receptors were within the urothelium middle cell layers and detrusor smooth muscle cells. The expression pattern of each 5-HT receptor subtype mRNA within the BPH urothelium was similar to that in the normal urothelium. The expression level of 5-HT2A receptor mRNA in the BPH group was significantly lower than the normal group; however, the expressions of both 5-HT3A and 5-HT7 mRNAs were significantly higher. The expressions of both 5-HT2B and 5-HT4 mRNAs were not significantly different between the normal and BPH groups. Conclusion: In normal urinary bladders, the expressions of both 5-HT2B and 5-HT7 mRNAs were higher compared to the 5-HT2A, 5-HT3A, and 5-HT4 mRNAs. The distributions of 5-HT2B and 5-HT7 receptors were different in the urothelium and detrusor layers. The 5-HT3A and 5-HT7 receptor mRNAs in the BPH group were significantly higher compared to the normal urothelium, while the 5-HT2A mRNA was significantly lower.ArticleADVANCES IN THERAPY.32:S29-S37(2015)journal articl

    Assisted reproductive technology in Japan : A summary report for 2016 by the Ethics Committee of the Japan Society of Obstetrics and Gynecology

    Purpose: The Japan Society of Obstetrics and Gynecology started an online cycle-based assisted reproductive technology (ART) registry system in 2007. This report presents the characteristics and treatment outcomes of ART registered for the cycles practiced during 2016. Methods: Cycle‐specific information for all ART cycles implemented in participating ART facilities were collected. A descriptive analysis was conducted for the registry database of 2016. Results: In total, 447 790 treatment cycles and 54 110 neonates (one in 18.1 neonates born in Japan) were reported in 2016. The mean patients’ age was 38.1 years (SD = 4.5). Among the egg retrieval cycles, 104 575 of 251 399 (41.6%) were freeze-all cycles without fresh embryo transfers (ET), while fresh ET was performed in 64 497 cycles (58.4%). A total of 187 132 frozen‐thawed ET cycles were reported, resulting in 62 432 pregnancies and 44 484 neonates born. Single ET was selected for 81.0% of fresh transfers and 82.7% of frozen cycles, resulting in singleton pregnancy/live birth rates of 97.0%/96.4% and 96.7%/96.4%, respectively. Conclusion: The total ART cycles and subsequent live births continued to increase in 2016. Single ET was performed more than 80%, and ET has shifted from using fresh embryos to frozen ones

    High-resolution melting curve analysis for rapid detection of mutations in a Medaka TILLING library

    <p>Abstract</p> <p>Background</p> <p>During the last two decades, DNA sequencing has led to the identification of numerous genes in key species; however, in most cases, their functions are still unknown. In this situation, reverse genetics is the most suitable method to assign function to a gene. TILLING (Targeting Induced Local Lesions IN Genomes) is a reverse-genetic strategy that combines random chemical mutagenesis with high-throughput discovery of the induced mutations in target genes. The method has been applied to a variety of plant and animal species. Screening of the induced mutations is the most important step in TILLING. Currently, direct sequencing or nuclease-mediated screening of heteroduplexes is widely used for detection of mutations in TILLING. Both methods are useful, but the costs are substantial and turnaround times are relatively long. Thus, there is a need for an alternative method that is of higher throughput and more cost effective.</p> <p>Results</p> <p>In this study, we developed a high resolution melting (HRM) assay and evaluated its effectiveness for screening ENU-induced mutations in a medaka TILLING library. We had previously screened mutations in the <it>p53 </it>gene by direct sequencing. Therefore, we first tested the efficiency of the HRM assay by screening mutations in <it>p53</it>, which indicated that the HRM assay is as useful as direct sequencing. Next, we screened mutations in the <it>atr </it>and <it>atm </it>genes with the HRM assay. Nonsense mutations were identified in each gene, and the phenotypes of these nonsense mutants confirmed their loss-of-function nature.</p> <p>Conclusions</p> <p>These results demonstrate that the HRM assay is useful for screening mutations in TILLING. Furthermore, the phenotype of the obtained mutants indicates that medaka is an excellent animal model for investigating genome stability and gene function, especially when combined with TILLING.</p

    Assisted reproductive technology in Japan : a summary report of 1992–2014 by the Ethics Committee, Japan Society of Obstetrics and Gynecology

    Aim: The Japan Society of Obstetrics and Gynecology implemented a registry report system for the clinical practice of assisted reproductive technology in 1986. The aggregated results from 1992 to 2014 are reported herein. Methods and Results: The total number of registered treatments was 393 745 cycles, of which 66 550 were pregnancy cycles and 46 008 were cycles with a live birth. Compared to the number of registered treatments in 2008, when the cycle‐based registry was newly introduced, there was a 2.07‐fold increase in the total number of treatments and a 2.25‐fold increase in the number of cycles with a live birth. As the average age of patients who receive assisted reproductive technology has become markedly higher year by year, the most common age of those patients who received assisted reproductive technology in 2014 was 40 years. Conclusion: The total numbers of both assisted reproductive technology treatments and assisted reproductive technology live births are likely to be higher in the future. In addition, the trend toward aging patients seems to be continuing into the future

    Revisiting PFA-mediated tissue fixation chemistry: FixEL enables trapping of small molecules in the brain to visualize their distribution changes

The spatial distribution, target selectivity, and diffusion/excretion kinetics of small molecules in structurally complicated specimens are critical for function. However, robust methods for precisely evaluating these parameters in the brain have been limited. Herein, we report a new method termed “fixation-driven chemical cross-linking of exogenous ligands (FixEL), ” which traps and images exogenously administered molecules of interest (MOIs) in complex tissues. This method relies on protein-MOI interactions and chemical cross-linking of amine-tethered MOI with paraformaldehyde used for perfusion fixation. FixEL is used to obtain images of the distribution of the small molecules, which addresses selective/nonselective binding to proteins, time-dependent localization changes, and diffusion/retention kinetics of MOIs such as the scaffold of PET tracer derivatives or drug-like small molecules

    Bioorthogonal chemical labeling of endogenous neurotransmitter receptors in living mouse brains

Because the spatiotemporal expression profiles and dynamics of neurotransmitter receptors involved in many functions are delicately governed in the brain, in vivo research tools with high spatiotemporal resolution for receptors in intact brains are highly desirable. Covalent labeling by chemical reaction (chemical labeling) of proteins without genetic manipulation is now a powerful method for analyzing receptors in vitro. However, selective target receptor labeling in the brain has not yet been achieved. This study shows that ligand-directed alkoxyacylimidazole (LDAI) chemistry can be used to selectively tether synthetic probes to target endogenous receptors in living mouse brains. The reactive LDAI reagents with negative charges were found to diffuse well over the whole brain and could selectively label target endogenous receptors, including AMPAR, NMDAR, mGlu1, and GABAAR. This simple and robust labeling protocol was then used for various applications: three-dimensional spatial mapping of endogenous receptors in the brains of healthy and disease-model mice; multi-color receptor imaging; and pulse–chase analysis of the receptor dynamics in postnatal mouse brains. Here, results demonstrated that bioorthogonal receptor modification in living animal brains may provide innovative molecular tools that contribute to the in-depth understanding of complicated brain functions

    Early gastric cancer detection in high-risk patients: a multicentre randomised controlled trial on the effect of second-generation narrow band imaging

    Objective: Early detection of gastric cancer has been the topic of major efforts in high prevalence areas. Whether advanced imaging methods, such as second-generation narrow band imaging (2G-NBI) can improve early detection, is unknown. Design: This open-label, randomised, controlled tandem trial was conducted in 13 hospitals. Patients at increased risk for gastric cancer were randomly assigned to primary white light imaging (WLI) followed by secondary 2G-NBI (WLI group: n=2258) and primary 2G-NBI followed by secondary WLI (2G-NBI group: n=2265) performed by the same examiner. Suspected early gastric cancer (EGC) lesions in both groups were biopsied. Primary endpoint was the rate of EGC patients in the primary examination. The main secondary endpoint was the positive predictive value (PPV) for EGC in suspicious lesions detected (primary examination). Results: The overall sensitivity of primary endoscopy for the detection of EGC in high-risk patients was only 75% and should be improved. 2G-NBI did not increase EGC detection rate over conventional WLI. The impact of a slightly better PPV of 2G-NBI has to be evaluated further. Trial registration number: UMIN000014503
